Описание: 2D HPLC is the selective transfer of a fraction or fractions from one column to another for further separation. It can be used in a variety of situations including:
- Further resolution of a complex mixture that can’t be separated on a single column (1D chromatography)
- Sample cleanup, removing matrix or interfering compounds
- Trace enrichment
In this presentation, we’ll talk about the approaches to 2D HPLC, comprehensive and heart-cutting (single and multiple). While using reversed phase in both dimensions is the most common combination for 2D HPLC, it has the lowest orthogonality. We’ll look at how combining orthogonal techniques, like IEX and RP can help you achieve optimum resolution and discuss the advantages and disadvantages of these combinations.
